At Bombi, we aim to solve some of today’s most difficult therapeutic challenges; fighting bacterial resistance while safeguarding the skin's microbiome while treating inflammatory skin disorders caused by bacterial infection.

Since the rise in multi-drug resistant skin infections seems unstoppable, alternative strategies to combat these pathogens are of high unmet need. Our platform is based on broad-spectrum antimicrobial peptides derived from the skin secretion of the Oriental fire-bellied toad (Bombina orientalis) which are then further conjugated to a bacterial recognition domain, also known as "species-specific quorum sensing peptide motifs" which can either be designed to target gram-negative or gram-positive bacteria, or even fungi.

The resulting linear peptides are further enhanced by N-terminus palmitoylation in order to provide conformational stability, binding potency/selectivity, and increase their antibacterial activity.

The killing moiety of the resulting fusion peptides are redesigned by modifying electrostatic attractions between cationic peptide molecules and anionic lipids of the bacterial membrane, thereby causing a discriminant and targeted bactericidal effect via a species-specific receptor-mediated pathway.

Our engineering strategy ultimately results in the generation of novel fusion peptides that are switched from being less-specific broad-spectrum to highly selective with increased potency and biofilm penetration properties.
